Project A01 – Drug-induced reprogramming of modified RNA nucleosides in the human RNome


Project A01 investigates how pharmaceutical compounds reprogram RNA and DNA modifications in human cells. Building on the mass spectrometry platforms developed in the first two funding periods, we apply nucleoside- and oligonucleotide-level MS to dissect drug-induced changes in tRNA, rRNA, and chromatin-associated RNA. We focus on modification loss under antimetabolite stress, compensatory methylation during queuine deficiency, and the epigenomic and epitranscriptomic effects of drugs linked to depression. In close collaboration with other subprojects, this work will provide mechanistic insights into RNA modification dynamics in the context of chemical biology.
