Project C05 – Fe2+/α-KG-dependent Enzymes and Biomimetics Involved in Epigenetics and DNA repair in extreme environments


We will characterize Fe2+/α-KG-dependent dioxygenases from extremophilic bacteria (such as Methylacidiphilum fumariolicum SolV) and through RNAseq-based gene expression analysis also characterize how radiation induced DNA damage is being repaired in these bacteria. Extremophiles, such as SolV that thrive at high temperatures and an acidic pH, hold the potential for the discovery of highly stable enzymes that can been used as tools in epigenetics. In addition, we plan on using the recently installed TIMS-TOF Mass Spectrometer for questions explored in this CRC, for example to analyze nucleoside isomers and new modifications. Lastly, we will use our optimized biomimetic and inorganic reagents as selective tools for modification of nucleosides.
